在药代动力学分析中发现体重会影响多奈单抗的清除率和分布容积，但由此产生的变化并不意味着需要调整剂量。

无需根据体重调整剂量

体重增加会增加多奈单抗的清除率和分布容积，从而降低总体暴露量。^1-3

体重对暴露的临床相关性预计很小，因为稳态下最大浓度的变化很小，但稳态谷浓度没有显著变化：

• 当从基于体重的剂量（在 1 期研究中）改为固定剂量（在 2 期和 3 期研究中）时，以及
• 在药代动力学模拟中。^1-3

这些结果表明，两种方法的性能相似，建议对多奈单抗进行固定剂量。   ^1,3

在 3 期双盲、安慰剂对照的 TRAILBLAZER-ALZ 2 研究中：

• 入组患者的体重指数 （BMI） 范围为 13.0 至 49.4 kg/m²，以及
• 平均 BMI 为 25.9 kg/m²。¹ 

在这项研究中，多奈单抗没有按体重给药。⁴
